- Benicio Del Toro is a huge fan of the original film, and remained attached to the remake ever since it was first announced in 2006 and passed through the hands of several directors.
- Talbot Manor is really Chatsworth House located in the Peak District of Derbyshire, England (though with some digital embellishments).
- The novelization of the script was written by multiple Bram Stoker Award-winning author Jonathan Maberry and is scheduled for release from Tor Books in 2010.
- In an uncredited cameo, the character of Max von Sydow says he purchased the silver-headed cane at Gevaudan "lifetimes ago". Gevaudan is famous for a series of attacks on humans that were believed to be caused by werewolf known as the beast of Gevaudan. The scene featuring von Sydow appears only on the director's cut.
- Was at first slated to hit cinemas in 2007, but difficulties to find a director made this release date impossible. Then a new release date was set for February 2009, then November 2009 and then finally February 2010.
- This is Benicio Del Toro's second time playing a "wolfman". His very first feature film role was that of a "dog-faced boy" in Pee Wee Herman's Big Top Pee-wee (which its music was also scored by Danny Elfman).
- CAMEO(Rick Baker): Makeup artist Rick Baker appears as the gypsy man who whistles prior to the werewolf attack on the gypsy camp
- Inspector Abberline is a fictionalization of an actual historical person. He was the Scotland Yard Inspector that investigated London's 'Jack the Ripper' murders in the late 1880s. His past is mentioned in the film.
- The "Ancient Gypsy Lore" text that Gwen Conliffe (Emily Blunt) is shown reading are the two pages "Luxorious - Luzan" from Encyclopedia Britannica.
- Danny Elfman was the original composer on the film and recorded a complete score inspired by Wojciech Kilar's score for Francis Ford Coppola's Bram Stoker's Dracula. His score was rejected as it did not fit the new tone of the film after several re-shoots and delays. Paul Haslinger was supposed to replace him, but Universal pulled the plug on that, and decided to re-instate Elfman's gothic score.
- This is the first film Joe Johnston has directed to be rated R.
- The Extended cut of the film begins with the Universal Logo that was being used in the 1940s, when the original came out.
- Mark Romanek was going to direct but left the project over budget disagreements.
- The bear owned by the gypsies is actually recycled animation from The Golden Compass. The Polar Bear was changed into a Grizzly for this film.
- The way Sir John plays the harmonica while walking through the halls of the asylum was Anthony Hopkins' idea.
- When Rick Baker, who became a makeup artist because of films like The Wolf Man, heard that this film was being made he was working on the Universal film Norbit, he went into an executive's office to tell them he had to do the makeup.
- Many actors, including Gene Simmons and David Lee Roth, were called in to do the Wolfman's howls.
- Costume designer, Milena Canonero based The Wolfman costume on the werewolf's (Oliver Reed) costume in The Curse of the Werewolf. Most noticeable between the two is the open shirt
- Early in pre-production, Canadian theatre actor 'Mason Shumaker' was considered for the role of Lawrence Talbot/Wolfman. However Johnston eventually thought that Shumaker was too young for the part, and the studio wanted to go with a more bankable name.
- Benicio Del Toro's Wolfman make-up took approximately 3 hours to apply and 1 hour to remove.
- Robert Rodriguez described the long hair Benicio Del Toro used to play Jackie Boy in Sin City as "Wolf Man hair".
- SPOILER: When The Wolfman is being chased in London he jumps off a building and lands on a policeman. This was accomplished with real actors and not CGI.
- SPOILER: During the gypsy massacre Lawrence (Benicio Del Toro) saves a woman searching frantically for her daughter. The daughter is named Maria after Maria Ouspenskaya who played Maleva in The Wolf Man and Frankenstein Meets the Wolf Man.
- SPOILER: The only makeup effect that Rick Baker did not have anything to do with was the feral mountain boy who bites Sir John. The reason for this is because he felt it was a mistake to show it the way seen in the film. Roz Abery designed that makeup.
- SPOILER: Lawrence Talbot's mother dies in the film and he is haunted by it. Benicio Del Toro's own mother died when he was nine.
- SPOILER: In this film Lawrence (Benicio Del Toro) kills Sir John (Anthony Hopkins). In the original The Wolf Man Sir John (Claude Rains) kills Lawrence (Lon Chaney Jr.).
